The AFB Rapid Culture, Urine test is a specialized pathology test used to detect Mycobacterium tuberculosis (MTB Complex) or non-tuberculous mycobacteria (NTM or MOTT) in urine samples. It helps identify genitourinary tuberculosis (TB of the kidneys, bladder, or urinary tract) and other mycobacterial infections more quickly and accurately than conventional culture methods.












Understanding AFB Rapid Culture, Urine
Genitourinary tuberculosis (GUTB) is one of the most common forms of extrapulmonary tuberculosis, affecting the kidneys, urinary tract, and reproductive organs. It often develops when Mycobacterium tuberculosis spreads from the lungs to the urinary system through the bloodstream. Patients may experience symptoms such as painful urination, blood in urine, lower abdominal pain, fever, night sweats, or unexplained weight loss. Since these symptoms can overlap with other urinary tract conditions, an accurate diagnosis becomes essential.
The AFB (Acid-Fast Bacilli) Rapid Culture by Bactec MGIT (Mycobacteria Growth Indicator Tube) test plays a key role in diagnosing genitourinary tuberculosis and related infections. It detects the presence of Mycobacterium tuberculosis (MTB Complex) as well as non-tuberculous mycobacteria (NTM or MOTT) in urine samples. Unlike traditional culture methods that may take weeks, the MGIT system uses a special liquid medium and an oxygen-sensitive fluorescent sensor to accelerate and enhance bacterial detection. This fluorescence-based technology enables faster and more sensitive identification of mycobacterial growth, allowing doctors to confirm TB or related infections at an early stage, initiate timely treatment, monitor treatment response, and prevent complications.
Doctors may recommend this test when there are symptoms suggestive of genitourinary tuberculosis. It may also be advised if you have a history of exposure to someone with TB, HIV infection, or other conditions that weaken your immune system. In addition, individuals with a previous diagnosis of tuberculosis may be recommended this test for monitoring during and after treatment. It can also aid in identifying potential drug resistance in the bacteria, enabling doctors to guide the most appropriate treatment plan.
For this test, the urine sample must preferably be the first-morning midstream urine (part of the urine that comes after the first and before the last stream). Collect the urine sample in a sealed and sterile screw-capped container provided by our sample collection professional. Ensure that the urethral area (from where the urine is passed) is clean and the container doesn't come in contact with your skin. Women are advised not to give the sample during the menstrual period unless prescribed.

What does AFB Rapid Culture, Urine measure?
The AFB Rapid Culture, Urine test measures the growth of mycobacteria in urine samples, specifically detecting Mycobacterium tuberculosis (MTB Complex) and non-tuberculous mycobacteria (NTM or MOTT). The MGIT system uses a nutrient-rich liquid medium and an oxygen-sensitive fluorescent sensor. As mycobacteria grow and consume oxygen, fluorescence increases, signaling bacterial presence.
A positive result indicates active infection with Mycobacterium tuberculosis or another mycobacterial species. The cultured bacteria can also be used for drug susceptibility testing to guide treatment. A negative result suggests no mycobacteria were detected in the urine, making TB or related infections unlikely. Your doctor may recommend additional tests, such as PCR, if infection is still suspected.
Compared to microscopy or traditional solid culture methods, the MGIT system provides faster detection and higher sensitivity.
