Platelet-derived growth factor receptor alpha (PDGFRA) gene is mutated in gastrointestinal stromal tumors (GISTs), the most common mesenchymal tumors of the gastrointestinal tract. Mutations in these genes lead to the activation of their corresponding proteins. PDGFRA is mutated in about 10% of cases. Additionally, PDGFRA is also mutated in a variety of other cancers, including melanoma, lung cancer, and sarcoma. The mutations in these genes are thought to play a similar role in these cancers as they do in GIST, by promoting cell growth and survival.
